Also known as:

Afamelanotide, CUV1647, NDP-α-MSH

Melanotan 1 (Afamelanotide) is a linear synthetic analog of α-melanocyte stimulating hormone (α-MSH), consisting of 13 amino acids. Unlike the cyclic Melanotan 2, MT-1 is a linear peptide with more selective melanocortin receptor activity.

MT-1 was developed to provide tanning effects through MC1R activation while minimizing off-target effects on other melanocortin receptors. It has progressed further in clinical development than MT-2, with regulatory approvals in some jurisdictions for specific conditions.

The peptide is particularly relevant to research on photoprotection and conditions involving defective melanin production.

Melanotan 1 acts primarily through MC1R:

  • MC1R Selectivity: More selective for melanocortin 1 receptor than MT-2.
  • Melanogenesis: Stimulates melanin production in melanocytes.
  • Eumelanin: Promotes production of darker, photoprotective eumelanin.
  • DNA Repair: May enhance UV-induced DNA damage repair mechanisms.
  • Reduced Side Effects: Lower affinity for MC3R/MC4R means fewer off-target effects.

MT-1's photoprotective properties have been clinically studied:

  • Erythropoietic Protoporphyria: Approved for EPP in some regions.
  • UV Tolerance: Increases time to sunburn in fair-skinned individuals.
  • DNA Protection: May enhance DNA repair after UV exposure.
  • Melanin Density: Increases protective melanin in skin.

Clinical trials have established its photoprotective efficacy.

MT-1 has advanced clinical development:

  • FDA Approval: Approved for erythropoietic protoporphyria.
  • Safety Profile: Well-characterized safety from clinical trials.
  • Vitiligo: Studied for repigmentation in vitiligo patients.
  • Polymorphous Light Eruption: Investigated for sun-sensitive skin conditions.

MT-1 is one of the few melanocortin peptides with regulatory approval.
